Overview

Riboxxim is a synthetic, well-defined double-stranded RNA immunostimulant that acts as a dual agonist of Toll-like receptor 3 (TLR3) and the cytosolic RNA sensor RIG-I, developed by Riboxx Pharmaceuticals as a GMP-grade immune adjuvant for cancer and antiviral immunotherapy.[1][3][5][7] It consists of a 100 base-pair dsRNA with a 5′-triphosphate moiety, enabling robust activation of TLR3 and RIG-I signaling, which drives strong type I interferon production, Th1-skewed immunity, and potent activation of dendritic cells, CD8+ T cells, and NK cells while exhibiting lower toxicity and better-defined structure than classical poly(I:C) analogues.[1][3][5] In preclinical models, Riboxxim, especially when encapsulated in PLGA particles together with tumor or viral antigens, elicits vigorous antigen-specific cytotoxic T-cell responses, retards primary tumor growth, prevents metastases, and synergizes with immune checkpoint blockade (e.g., anti-CTLA-4) to achieve durable tumor ablation, and has also been used as an intranasal adjuvant in experimental SARS-CoV-2 vaccines to induce mucosal and systemic immunity.[2][3][5][8]
