Ribozinoindole-2 (Rbin-2) is a potent, selective, and reversible small molecule inhibitor of midasin (MDN1), an essential AAA+ ATPase involved in eukaryotic ribosome biogenesis. Discovered by researchers at The Rockefeller University and The University of Tokyo, Rbin-2 directly targets midasin to inhibit its ATPase activity, thereby blocking the ATP-dependent removal of assembly factors from pre-60S ribosomal particles and disrupting large ribosomal subunit maturation. In preclinical oncology research, Rbin-2 has been investigated for its ability to target cancer stem cells (CSCs) and reverse endocrine resistance in estrogen receptor-positive (ER+) and ESR1-mutant breast cancers, where midasin is often overexpressed.
