RIDE-001 is a first-in-class ApoA1-based nanomedicine being developed by Trained Therapeutix Discovery to induce “trained immunity” in the innate immune system for the treatment of solid tumors. It is designed to selectively target bone marrow myeloid progenitor cells, where it activates the intracellular pattern recognition receptor NOD2, triggering epigenetic reprogramming that yields trained myeloid cells with enhanced antitumor activity.[1][4][6][13] In preclinical studies across mice, rats, and non-human primates, RIDE-001 demonstrated dose-dependent NOD2 activation, durable myeloid cell training, single-agent antitumor efficacy in immunosuppressive melanoma models, and synergy with anti-PD-1 checkpoint blockade in colorectal cancer models, along with a favorable safety profile.[1][4][6] The program is currently in the preclinical stage, with GMP manufacturing initiated in anticipation of first-in-human studies in patients with solid tumors.[1][4]
