Overview

Rifampicin + isoniazid + pyrazinamide is a fixed-dose combination oral medication used for the treatment of tuberculosis (TB). It combines three first-line anti-tuberculosis agents: - **Rifampicin** (also known as rifampin), a rifamycin antibiotic that inhibits bacterial DNA-dependent RNA polymerase, thereby suppressing RNA synthesis and leading to bacterial cell death. - **Isoniazid**, which inhibits the synthesis of mycolic acids essential for the mycobacterial cell wall. - **Pyrazinamide**, which disrupts membrane transport and energy production in Mycobacterium tuberculosis under acidic conditions. This combination targets Mycobacterium tuberculosis by attacking different aspects of its biology, reducing the risk of resistance development and improving treatment adherence. The fixed-dose formulation simplifies therapy by reducing pill burden. It is typically used during the intensive phase of TB treatment, often followed by continuation with other drugs[1][3][5][7][8].
