Overview
This is a combination therapy of recombinant interleukin-21 (rIL-21), a cytokine with immunostimulatory properties, and sorafenib, an oral multikinase inhibitor. rIL-21 acts as an immune modulator by stimulating the proliferation and activity of cytotoxic T cells and natural killer (NK) cells. Sorafenib inhibits multiple intracellular serine/threonine kinases in the Ras/mitogen-activated protein kinase (MAPK) pathway—including Raf isoforms—and cell surface tyrosine kinase receptors such as KIT, FLT3, RET/PTC, VEGFRs 1–3, and PDGFR-beta. The combination has been studied in metastatic renal cell carcinoma (mRCC), where it demonstrated antitumor activity and acceptable safety in previously treated patients[2][4].
