Overview

Rilonacept is a dimeric fusion protein biologic that acts as an interleukin-1 (IL-1) inhibitor. It is composed of portions of the human interleukin-1 receptor (IL-1R) and the IL-1 receptor accessory protein, fused to the Fc portion of human IgG1. Rilonacept binds and neutralizes both IL-1α and IL-1β, preventing their interaction with cell surface receptors and thereby inhibiting downstream inflammatory signaling. It is primarily indicated for the treatment of cryopyrin-associated periodic syndromes (CAPS), including familial cold autoinflammatory syndrome (FCAS) and Muckle-Wells syndrome (MWS), in adults and children 12 years or older; for maintenance of remission in deficiency of interleukin-1 receptor antagonist (DIRA) in patients weighing at least 10 kg; and for treatment and prevention of recurrent pericarditis in adults and children 12 years or older[2][3][4][5][6][7].
