Overview

Rilotumumab is a fully human monoclonal antibody that targets hepatocyte growth factor (HGF), the only known ligand of the MET receptor. By binding to HGF, rilotumumab blocks its interaction with the MET receptor, thereby inhibiting downstream signaling pathways involved in cell proliferation, survival, motility, and invasion. The drug was developed for the treatment of various solid tumors, most notably advanced gastric or gastroesophageal junction adenocarcinoma. Rilotumumab was developed by Amgen and underwent extensive clinical evaluation; however, all clinical trials were halted after increased mortality was observed in patients receiving rilotumumab plus chemotherapy compared to chemotherapy alone[1][3][4][6].
