Overview
A combination therapy consisting of rilotumumab (a fully human monoclonal antibody that neutralizes hepatocyte growth factor, HGF), epirubicin (an anthracycline topoisomerase II inhibitor), cisplatin (a platinum-based DNA-damaging agent), and capecitabine (an oral prodrug converted to 5-fluorouracil, an antimetabolite). This combination was investigated as a first-line treatment in advanced, MET-positive gastric or gastro-esophageal junction adenocarcinoma. Rilotumumab interrupts HGF/MET signaling implicated in tumor growth and metastasis. Clinical trials were discontinued early due to higher mortality in the rilotumumab arm[1][3][5].
