rilvegostomig + S-1 is a combination therapy being developed by AstraZeneca for the treatment of biliary tract cancer. It consists of rilvegostomig (AZD2936), a bispecific monoclonal antibody that simultaneously targets the PD-1 and TIGIT immune checkpoints, and S-1, an oral fluoropyrimidine chemotherapy agent. S-1 itself is a multi-component drug containing tegafur (a 5-fluorouracil prodrug), gimeracil (a dihydropyrimidine dehydrogenase inhibitor), and oteracil (an orotate phosphoribosyltransferase inhibitor). The regimen is currently being evaluated in the Phase III ARTEMIDE-Biliary01 trial as an adjuvant treatment for patients with resected biliary tract cancer who are at high risk of recurrence. By combining immune checkpoint blockade with cytotoxic chemotherapy, the treatment aims to enhance the immune system's ability to target cancer cells and eliminate residual disease following surgical resection.
