Overview

Rimiducid is a lipid-permeable tacrolimus analogue and small molecule protein dimerizer designed to enhance the control of immune cell activity in cellular immunotherapies for cancer and blood disorders. It acts as a homodimerizing agent that binds to engineered drug-binding domains derived from human FK506-binding protein (FKBP12), specifically those with the F36V mutation, present on modified proteins such as inducible caspase-9. Upon administration, rimiducid induces dimerization and activation of these switch proteins—most notably activating caspase-9—which triggers apoptosis in genetically modified T-cells. This mechanism is used as a "safety switch" in CAR T-cell therapies to rapidly eliminate problematic or overactive cells (such as during graft-versus-host disease) while sparing non-alloreactive T-cells. Rimiducid is under investigation primarily for use alongside cellular immunotherapies in hematological cancers and other conditions where precise control of immune cell populations is required[1][2][3][6].
