Overview

RiMO-301 is a hafnium-based metal-organic framework (MOF) compound developed as a radio-enhancer for cancer therapy. Upon intratumoral administration and subsequent irradiation of the tumor site, RiMO-301 absorbs X-ray photons and generates reactive oxygen species (ROS), such as hydroxyl radicals and singlet oxygen. These ROS induce DNA damage in irradiated cancer cells, leading to tumor cell lysis. Preclinical models have shown that RiMO-301 can enhance the effects of radiotherapy and checkpoint blockade immunotherapy without significant systemic toxicity. Clinical studies have demonstrated promising efficacy in patients with advanced tumors, particularly head and neck cancers, when used with or without pembrolizumab during palliative radiotherapy[1][2][3].
