The Chemokine Modulatory Regimen (CKM) is an investigational combination immunotherapy consisting of rintatolimod (a selective TLR3 agonist), interferon alfa-2b (a cytokine), and celecoxib (a COX-2 inhibitor). Developed at Roswell Park Comprehensive Cancer Center, the regimen is designed to reprogram the tumor microenvironment by inducing the production of T-cell-attracting chemokines such as CXCL9, CXCL10, and CCL5. This process aims to convert immunologically "cold" tumors into "hot" tumors by increasing the infiltration of cytotoxic T lymphocytes (CTLs), thereby enhancing the efficacy of other immunotherapies or the body's natural anti-tumor response. It has been primarily evaluated in Phase 1 and Phase 2 clinical trials for colorectal cancer, breast cancer, and melanoma.
