Overview

The Chemokine Modulatory Regimen (CKM) is an investigational combination immunotherapy developed at Roswell Park Comprehensive Cancer Center. It consists of three active components: rintatolimod (a selective TLR3 agonist), interferon alfa-2b (a cytokine), and celecoxib (a COX-2 inhibitor). The regimen is designed to reprogram the tumor microenvironment by selectively inducing the production of cytotoxic T lymphocyte (CTL)-attracting chemokines, such as CXCL9, CXCL10, and CCL5. By increasing the infiltration of CD8+ T cells, CKM aims to convert immunologically "cold" tumors into "hot" tumors, potentially enhancing the efficacy of other immunotherapies. It has been evaluated in Phase 1 and 2 clinical trials for various solid tumors, including colorectal cancer, triple-negative breast cancer, and melanoma.
