RIP140 small interfering RNA is an RNA interference (RNAi)-based therapeutic candidate developed by CytRx R&D Company for the treatment of obesity and type 2 diabetes. The drug utilizes small interfering RNA (siRNA) to target and silence the expression of Receptor-interacting protein 140 (RIP140), also known as Nuclear receptor-interacting protein 1 (NRIP1). RIP140 is a transcriptional corepressor that normally inhibits metabolic pathways; its depletion leads to the upregulation of genes involved in glucose uptake, glycolysis, fatty acid oxidation, and mitochondrial biogenesis. In preclinical models, this mechanism has demonstrated the ability to increase metabolic rate and provide resistance to diet-induced obesity.
