Overview

The RIPK2 program by Atomwise (now Numerion Labs) focuses on the development of small molecule scaffolding inhibitors targeting Receptor-interacting protein-kinase 2 (RIPK2). Unlike traditional kinase inhibitors that target catalytic activity, these molecules are designed to disrupt the protein-protein interactions (scaffolding) of RIPK2. This mechanism aims to inhibit the NOD1 and NOD2 signaling pathways, which are critical mediators of the innate immune response and chronic inflammation. The program includes both peripheral candidates for inflammatory bowel disease (IBD) and CNS-penetrant molecules for neuroinflammatory diseases.
