Drug intelligence / Profile preview
RIPR-PD-1 (Receptor Inhibition by Phosphatase Recruitment PD-1) is a bispecific molecule designed to attenuate programmed cell death protein 1 (PD-1) signaling. Unlike conventional checkpoint inhibitors that block ligand binding, RIPR-PD-1 forces the cis-ligation of PD-1 to the abundant cell-surface phosphatase CD45 (receptor-type tyrosine-protein phosphatase C). This proximity-induced recruitment of CD45 leads to the direct intracellular dephosphorylation of tyrosine residues on the PD-1 receptor, thereby inhibiting both ligand-activated and ligand-independent tonic PD-1 signaling. RIPR-PD-1 has demonstrated enhanced T-cell and dendritic cell activation and superior anti-tumor efficacy compared to standard anti-PD-1 antibodies in preclinical models.
