Overview
Risperidone and olanzapine are second-generation (atypical) antipsychotics used primarily for the treatment of schizophrenia and bipolar disorder. In the context of clinical research for treatment-resistant schizophrenia (TRS), such as the Lu AF35700 'DayBreak' trial, they are frequently utilized as the 'conventional therapy' or active comparator. Risperidone is a benzisoxazole derivative that acts as a potent D2 and 5-HT2A receptor antagonist. Olanzapine is a thienobenzodiazepine with a broader pharmacological profile, antagonizing D2, 5-HT2A, H1, M1, and alpha-1 receptors. Both agents are designed to provide antipsychotic efficacy with a lower incidence of extrapyramidal symptoms compared to first-generation antipsychotics.
