Risperidone + olanzapine + aripiprazole refers to a set of three atypical antipsychotic medications used as standard-of-care treatments in a clinical research program led by Peking University. This program focuses on identifying characteristics and developing prediction models for treatment-resistant schizophrenia (TRS), specifically patients who are treatment-resistant from the illness onset (TRO). In the associated clinical trial, first-episode schizophrenia patients are randomly assigned to receive one of these three medications for a one-year period. Risperidone and olanzapine function primarily as antagonists at dopamine D2 and serotonin 5-HT2A receptors, while aripiprazole acts as a partial agonist at D2 and 5-HT1A receptors and an antagonist at 5-HT2A receptors. These drugs are well-established pharmaceuticals used to manage positive and negative symptoms of schizophrenia, and the Peking University study aims to use clinical and neurocognitive data from patients treated with these agents to improve early diagnosis of treatment resistance.
