Risuteganib + bevacizumab is a combination therapy used by intravitreal injection primarily for the treatment of diabetic macular edema (DME) and is also being investigated for age-related macular degeneration (AMD). Risuteganib (Luminate, ALG-1001) is a synthetic RGD-class peptide that acts as an integrin regulator, inhibiting multiple pathogenic pathways including angiogenesis, inflammation, and cell death; it exerts cytoprotective, anti-inflammatory, anti-angiogenic, and pro-mitochondrial effects in retinal cells. Bevacizumab (Avastin) is a recombinant monoclonal antibody that selectively binds and neutralizes vascular endothelial growth factor A (VEGF-A), thereby inhibiting angiogenesis and vascular permeability. This combination is explored in clinical trials to target both integrin- and VEGF-mediated pathways for enhanced efficacy, particularly in patients with DME who do not optimally respond to anti-VEGF monotherapy. Clinical trial data suggest the combination is non-inferior in improving best corrected visual acuity and reducing central macular thickness when compared to bevacizumab alone, with some evidence of increased durability of effect[1][5][3].
