Risvodetinib (IKT-148009) is a potent and selective small molecule inhibitor of Abelson tyrosine kinase (c-Abl), designed to block the activation of c-Abl and its closely related enzyme Abl2/Arg with high specificity and limited inhibition of other kinases such as c-Kit or PDGFRa/b. It has nearly 25 times the potency of imatinib against c-Abl in enzyme inhibition assays. The drug is being developed by Inhibikase Therapeutics for neurodegenerative diseases, primarily Parkinson's disease and multiple system atrophy (MSA). Risvodetinib acts by inhibiting c-Abl in neurons where alpha-synuclein has activated the enzyme, thereby suppressing neurodegeneration. Preclinical studies have demonstrated its neuroprotective properties even when treatment was delayed after disease onset. Clinical trials have shown that risvodetinib is well tolerated with good central nervous system penetration[3][4][6][8]. The drug has received Orphan Drug Designation from the FDA for MSA[7].
