Drug intelligence / Profile preview
Risvutatug (also referred to by the machine-translated name livototamab) is a fully human monoclonal antibody targeting B7-H3 (CD276), an immune checkpoint ligand overexpressed in various solid tumors. It is developed as the antibody-drug conjugate (ADC) risvutatug rezetecan (HS-20093 / GSK5764227), where risvutatug is covalently linked to rezetecan, an exatecan-derived topoisomerase I inhibitor payload, via a cleavable tetrapeptide linker. Upon binding to B7-H3 on tumor cells, the ADC is internalized, releasing the topoisomerase I inhibitor to induce DNA damage and apoptosis. Originally developed by Hansoh Pharma and licensed globally (excluding Greater China) to GSK, risvutatug rezetecan is in Phase III clinical trials for small cell lung cancer (SCLC) and osteosarcoma, and in Phase II trials for other solid tumors including prostate cancer and non-small cell lung cancer.
