**Risvutatug rezetecan** is a novel B7-H3-targeted antibody-drug conjugate (ADC) composed of a fully human anti-B7-H3 monoclonal antibody covalently linked to a topoisomerase I inhibitor (TOP1i) payload. It selectively binds to B7-H3, a cell surface protein overexpressed in various solid tumors, enabling targeted delivery of the cytotoxic payload to induce DNA damage and tumor cell death. Developed originally by Hansoh Pharmaceutical with exclusive worldwide rights (ex-China, Hong Kong, Macau, Taiwan) licensed to GSK, it has shown promising antitumor activity in heavily pretreated patients across multiple solid tumors, including high response rates in small cell lung cancer (SCLC) and osteosarcoma, with a manageable safety profile primarily featuring hematologic toxicities.[1][3][7]
