Overview

ritlecitinib + narrowband UVB is a combination therapeutic regimen being investigated for the treatment of non-segmental vitiligo (NSV). Ritlecitinib is an oral small molecule that acts as a selective dual inhibitor of Janus kinase 3 (JAK3) and the tyrosine kinase expressed in hepatocellular carcinoma (TEC) family kinases. By inhibiting these pathways, ritlecitinib blocks the signaling of cytokines such as IL-15 and the activity of CD8+ T-cells, which are primary drivers of melanocyte destruction in vitiligo. Narrowband UVB (nbUVB) is a form of phototherapy that utilizes a specific wavelength of ultraviolet radiation (311–312 nm) to stimulate the differentiation and proliferation of melanocyte stem cells, facilitating repigmentation. The combination approach aims to simultaneously arrest the autoimmune process and promote the regeneration of pigment-producing cells. This specific combination is currently being evaluated in a Phase 2 multicentric study led by the Centre Hospitalier Universitaire de Nice.
