This is a combination regimen consisting of three agents: rituximab, bortezomib, and Y-90 ibritumomab tiuxetan. Rituximab is a monoclonal antibody targeting the CD20 antigen on B lymphocytes, leading to cell lysis primarily via complement-dependent cytotoxicity and antibody-dependent cellular cytotoxicity. Bortezomib is a small molecule proteasome inhibitor that disrupts protein degradation pathways in cancer cells, resulting in apoptosis. Y-90 ibritumomab tiuxetan (Zevalin) is a radioimmunoconjugate composed of an anti-CD20 monoclonal antibody (ibritumomab) linked to the radioactive isotope yttrium-90; it delivers targeted radiation to CD20-positive B-cells, inducing cell death through both immunologic and radiologic mechanisms. This combination has been investigated for relapsed or refractory follicular non-Hodgkin lymphoma and other B-cell lymphomas[1][2][3][4][5].
