Overview

This combination is a chemotherapy regimen primarily used for the treatment of B-cell non-Hodgkin lymphomas, such as diffuse large B-cell lymphoma (DLBCL). It is essentially the standard R-CHOP regimen with the omission of vincristine, often to avoid neurotoxicity in susceptible patients or as a backbone for investigational agents. The regimen consists of rituximab, a monoclonal antibody targeting CD20; cyclophosphamide, an alkylating agent; doxorubicin, an anthracycline topoisomerase II inhibitor; and prednisone, a corticosteroid. Together, these agents work synergistically to induce B-cell depletion and DNA damage-mediated apoptosis in malignant lymphocytes.
