Overview

Rituximab + DC is an investigational combination cancer vaccine regimen developed by Oslo University Hospital. It consists of the anti-CD20 monoclonal antibody rituximab and autologous mature dendritic cells (DC) derived from the patient's own leukapheresed cells. The regimen is administered via intratumoral injection into a lymph node following local low-dose radiotherapy. This approach functions as an "in situ" vaccine: radiotherapy induces tumor cell death and the release of antigens, while rituximab opsonizes tumor cells to facilitate their uptake via Fc receptors on the injected dendritic cells. These dendritic cells then process and present the tumor antigens to the systemic immune system, intended to prime a T-cell-mediated anti-tumor response. The regimen has been primarily studied in Phase II trials for patients with untreated or relapsed indolent non-Hodgkin's lymphoma (iNHL), including follicular lymphoma.
