Overview

Rituximab + fludarabine + cyclophosphamide (RFC) is a combination chemoimmunotherapy regimen used primarily in the treatment of B-cell malignancies, such as follicular lymphoma and chronic lymphocytic leukemia. The regimen consists of **rituximab**, a chimeric monoclonal antibody that targets the CD20 antigen on B-lymphocytes; **fludarabine**, a purine nucleoside analog that interferes with DNA synthesis; and **cyclophosphamide**, an alkylating agent that cross-links DNA. This specific combination was evaluated by the **Asociacion Espanola de Hematologia y Hemoterapia (AEHH)** in the Phase 4 LNHF-03 clinical trial as a first-line treatment for follicular lymphoma, followed by rituximab maintenance. While the regimen demonstrated high efficacy with significant complete remission rates, it is also associated with notable hematological toxicity.
