Overview

Rituximab + gemcitabine + cisplatin + methylprednisolone is a chemotherapy regimen primarily investigated as a salvage therapy for patients with relapsed or refractory diffuse large B-cell lymphoma (DLBCL)[1][5]. The regimen combines components with distinct mechanisms: rituximab (a monoclonal antibody targeting CD20 on B cells), gemcitabine and cisplatin (chemotherapeutic agents that damage DNA and inhibit replication in rapidly dividing cells), and methylprednisolone (a synthetic corticosteroid with anti-inflammatory and lympholytic effects)[1][3]. The combination is notable for limited overlapping toxicity with first-line therapies and is deliverable as a short inpatient regimen[5]. In clinical studies, adding rituximab to the backbone of gemcitabine, cisplatin, and methylprednisolone (GEM-P) resulted in improved response rates and progression-free survival compared to GEM-P alone[1][5].
