Triple combination therapy consisting of the anti-CD20 monoclonal antibody rituximab, the PI3K delta inhibitor idelalisib, and the BCL-2 inhibitor venetoclax. It is being evaluated in the Phase 1 RIVe-CLL/SLL trial (NCT03639324) led by Virginia Commonwealth University for the treatment of relapsed or refractory chronic lymphocytic leukemia (CLL) and small lymphocytic lymphoma (SLL). Dose combination 1-3 specifically refers to a regimen of 100 mg BID of idelalisib and 200 mg QD of venetoclax, administered following a lead-in period with idelalisib and rituximab. The combination aims to achieve deeper clinical responses by targeting multiple survival pathways in B-cell malignancies.
