Overview

This is a combination regimen consisting of four agents: - **Rituximab** is a monoclonal antibody targeting CD20 on B cells, leading to their depletion. It is widely used in the treatment of B-cell lymphomas. - **Inotuzumab ozogamicin** (previously known as CMC544) is an antibody-drug conjugate that targets CD22 on B cells and delivers a cytotoxic agent (calicheamicin), resulting in cell death. - **Gemcitabine** is a nucleoside analog (antimetabolite) that inhibits DNA synthesis and induces apoptosis. - **Oxaliplatin** is a platinum-based chemotherapeutic agent that causes DNA crosslinking and apoptosis. The combination has been studied for the treatment of relapsed or refractory diffuse large B-cell lymphoma (DLBCL), particularly in patients not eligible for high-dose chemotherapy or autologous stem cell transplantation. The rationale for this regimen includes synergistic anti-tumor effects from targeting multiple pathways involved in lymphoma survival and proliferation[1][2][8].
