R-PMDT is an investigational chemoimmunotherapy combination regimen designed for the treatment of newly diagnosed primary central nervous system lymphoma (PCNSL). The regimen integrates five therapeutic agents: rituximab (an anti-CD20 monoclonal antibody), pirtobrutinib (a highly selective, non-covalent Bruton's tyrosine kinase inhibitor), high-dose methotrexate (an antimetabolite), dexamethasone (a corticosteroid), and thiotepa (an alkylating agent). This combination is specifically structured to leverage agents with known efficacy in B-cell malignancies and the ability to penetrate the blood-brain barrier, aiming to improve the overall response rate and survival outcomes in patients with PCNSL.
