This regimen combines **rituximab** (a monoclonal antibody targeting CD20, leading to B-cell depletion), **polatuzumab vedotin** (an antibody-drug conjugate targeting CD79b, which delivers monomethyl auristatin E (MMAE) to B cells and disrupts microtubules, causing apoptosis), and **venetoclax** (an orally bioavailable small-molecule inhibitor of BCL-2 that triggers apoptosis in BCL-2 dependent malignant cells). The combination is being investigated primarily in relapsed/refractory (R/R) diffuse large B-cell lymphoma (DLBCL) and follicular lymphoma, and also in mantle cell lymphoma[1][3][5][7][9]. It is administered intravenously (rituximab, polatuzumab vedotin) and orally (venetoclax). The regimen is not marketed under a unique brand name, being used in clinical trials for high-risk, heavily pre-treated B-cell lymphomas.
