Overview

Rituximab + temozolomide is a combination therapy used primarily as a salvage regimen for recurrent and refractory primary central nervous system lymphoma (PCNSL). Rituximab is a chimeric monoclonal antibody that targets the CD20 antigen on B lymphocytes, leading to cell lysis through complement-dependent cytotoxicity and antibody-dependent cell-mediated cytotoxicity. Temozolomide is an oral alkylating agent that crosses the blood-brain barrier and induces DNA damage by methylating guanine residues at the O6 and N7 positions, resulting in apoptosis of tumor cells. The combination leverages rituximab’s immunotherapeutic effects with temozolomide’s cytotoxic activity to enhance anti-lymphoma efficacy[1][2][3][4][5][6].
