Riva Therapeutics is developing a small molecule inhibitor program targeting DNA polymerase theta (Polθ, also known as POLQ). Polθ is a specialized DNA polymerase that plays a central role in the microhomology-mediated end joining (MMEJ) DNA repair pathway. This pathway is often utilized by cancer cells that are deficient in homologous recombination (HR), such as those with BRCA1 or BRCA2 mutations. Inhibition of Polθ in these HR-deficient cells leads to genomic instability and cell death through a synthetic lethal mechanism. The program is currently in the preclinical stage, aimed at treating various HR-deficient solid tumors, including breast, ovarian, and prostate cancers.
