Overview

Rivoceranib + paclitaxel is a combination therapy consisting of rivoceranib, an oral small-molecule tyrosine kinase inhibitor that selectively inhibits vascular endothelial growth factor receptor 2 (VEGFR-2), and paclitaxel, a microtubule-stabilizing chemotherapeutic agent. Rivoceranib blocks VEGFR-2-mediated signaling pathways, thereby inhibiting tumor angiogenesis and growth. Paclitaxel disrupts microtubule dynamics, leading to cell cycle arrest and apoptosis in rapidly dividing cancer cells. This combination has been investigated primarily for the treatment of advanced gastric or gastroesophageal junction (GEJ) cancer as second-line therapy, as well as in gastrointestinal stromal tumors (GIST) with high P-glycoprotein expression after failure of standard therapies. Clinical studies have shown manageable safety profiles and promising efficacy signals for this regimen[1][5].
