Rizatriptan + acetaminophen is a combination therapy used for the acute treatment of migraine headaches. Rizatriptan is a selective serotonin 5-HT1B/1D receptor agonist (triptan) that works by narrowing blood vessels in the brain, inhibiting pain signal transmission, and blocking the release of substances that cause migraine symptoms. Acetaminophen is an analgesic and antipyretic agent commonly used to relieve mild to moderate pain and reduce fever. The rationale for combining these agents lies in their complementary mechanisms—rizatriptan targets central neurovascular pathways specific to migraines, while acetaminophen provides general analgesia through central inhibition of prostaglandin synthesis. Clinical studies have shown that coadministration can be more effective than acetaminophen or placebo alone for rapid pain relief in acute migraine attacks[5]. This combination has not demonstrated clear superiority over rizatriptan monotherapy but may offer benefit as part of multi-mechanism therapy.
