RJx-01 is a proprietary, orally administered combination drug comprised of metformin and galantamine. Developed by Rejuvenate Biomed, it is being investigated as a treatment for sarcopenia—a progressive loss of skeletal muscle mass and strength that occurs with aging or disuse—and muscle weakness associated with chronic obstructive pulmonary disease (COPD). Both constituent drugs are already approved for other indications (metformin for type 2 diabetes; galantamine for Alzheimer’s disease), but their combination in RJx-01 represents a novel therapeutic approach. Preclinical studies demonstrated that RJx-01 improves physical performance, muscle mass and force, neuromuscular junction stability, mitochondrial health, autophagy, lysosomal function, satellite cell content, and reduces systemic inflammation in animal models of sarcopenia. In Phase 1b clinical trials involving elderly men with disuse-induced muscle atrophy (immobilization), RJx-01 was safe and well tolerated while significantly improving muscle strength recovery compared to placebo. The drug targets several cellular pathways implicated in age-related decline in muscle quality—including mitochondrial function and neuromuscular junction integrity—making it a first-in-class candidate for these indications[2][5][7][8][10].
