RK-33 is a first-in-class small molecule inhibitor of the DEAD-box RNA helicase DDX3X. Developed primarily by researchers at Johns Hopkins University, RK-33 binds to the ATP-binding pocket of DDX3X, thereby inhibiting its helicase activity. DDX3X is often overexpressed in various malignancies and plays a critical role in translation initiation, cell cycle progression, and DNA damage repair. By inhibiting DDX3X, RK-33 induces cell cycle arrest, promotes apoptosis, and sensitizes cancer cells to radiation and chemotherapy. It has shown preclinical efficacy across a range of cancers, including lung, breast, prostate, colorectal, and endometrial cancers.
