RLA01 is a single-stranded DNA (ssDNA) aptamer identified through cell-SELEX (Systematic Evolution of Ligands by Exponential Enrichment) for its high affinity and specificity toward epithelial ovarian cancer (EOC) cells. It functions as a targeting ligand that recognizes and binds to surface receptors overexpressed on EOC cells, such as the Caov-3 line, subsequently triggering receptor-mediated endocytosis. In preclinical research, RLA01 is frequently utilized to functionalize drug delivery vehicles, such as poly(lactic-co-glycolic acid) (PLGA) nanoparticles, to selectively deliver cytotoxic payloads like paclitaxel directly into tumor cells. This targeted approach aims to enhance the therapeutic index of chemotherapy by increasing intratumoral drug concentration while minimizing cellular uptake in non-target tissues, such as human ovarian surface epithelial (HOSE) cells.
