RLIP antisense (also known as R508) is an antisense oligonucleotide (ASO) therapeutic agent designed to target and deplete the expression of the Ral-interacting protein (RLIP, also known as RLIP76 or RalBP1). RLIP is a multifunctional membrane transporter protein that plays a critical role in the efflux of glutathione conjugates, clathrin-dependent endocytosis, and the mediation of multi-drug resistance in various cancers. By reducing RLIP levels, the therapeutic induces apoptosis in cancer cells through the inhibition of the JAK2/STAT3 signaling pathway and the subsequent downregulation of anti-apoptotic proteins such as Mcl-1 and Bcl-2. Preclinical studies have demonstrated its efficacy in inducing tumor regression across a wide range of malignancies, including melanoma, ovarian, lung, and breast cancers. In animal models, the treatment has shown minimal toxicity to normal tissues, with the most notable side effects being moderate hypoglycemia and hypolipidemia.
