Overview

RM175 is an organometallic ruthenium(II) arene complex developed as a potential antineoplastic agent. It consists of a ruthenium center coordinated to a hydrophobic tetrahydroanthracene arene ligand, a bidentate ethylenediamine ligand, and a chloride leaving group. RM175 demonstrates significant in vitro and in vivo activity against human ovarian cancer models, including those resistant to cisplatin and carboplatin. Its mechanism of action involves DNA binding, but unlike platinum-based drugs, it maintains efficacy in mismatch repair (MLH1) deficient cells, suggesting a novel mechanism that avoids common cross-resistance pathways.
