Overview

RmAb38E2-scFv8D3 is a brain-penetrant bispecific antibody designed for the treatment of Parkinson's disease and other alpha-synucleinopathies. It is composed of RmAb38E2, a monoclonal antibody with high selectivity for alpha-synuclein (αSYN) oligomers, fused to a single-chain variable fragment (scFv) of the 8D3 antibody, which targets the transferrin receptor (TfR). This design enables the antibody to cross the blood-brain barrier (BBB) via TfR-mediated transcytosis, significantly increasing its concentration within the central nervous system compared to standard monospecific antibodies. Preclinical studies in transgenic mouse models (L61) have shown that RmAb38E2-scFv8D3 reduces brain αSYN oligomer levels and promotes microglial activation, potentially offering a more effective immunotherapy for neurodegenerative diseases characterized by αSYN aggregation.
