RmAbSynO2 is a recombinant monoclonal antibody that specifically targets aggregated forms of alpha-synuclein (αSYN), a protein whose accumulation is a hallmark of Parkinson's disease and other synucleinopathies. Developed primarily as a research tool and a therapeutic lead, RmAbSynO2 aims to reduce the burden of pathological αSYN aggregates in the central nervous system. Because large antibodies typically have poor blood-brain barrier penetration, RmAbSynO2 has been used as the binding domain in the engineering of bispecific antibodies (e.g., RmAbSynO2-scFv8D3) that incorporate a transferrin receptor (TfR) shuttle to facilitate receptor-mediated transcytosis into the brain. Preclinical studies in transgenic mouse models have demonstrated that while the unmodified RmAbSynO2 has limited brain uptake compared to its bispecific counterparts, it remains a potent binder of TBS-T soluble aggregated αSYN.
