RMC-8839 is a potent, oral, and selective small molecule inhibitor targeting the KRAS G13C mutant protein. It was developed by Revolution Medicines as part of their RAS(ON) mutant-selective inhibitor pipeline. The drug is designed to directly inhibit the active (GTP-bound) form of KRAS with a G13C mutation, which is implicated in driving certain cancers. Preclinical data demonstrated that selective inhibition of active KRAS G13C with RMC-8839 reveals an increased dependence of codon 13 KRAS-mutant cancers on this oncogenic driver[5][9]. However, development has been discontinued and it did not advance to later clinical phases[3][4].
