RMC-9945 is a potent and selective small-molecule inhibitor of the active, GTP-bound (ON) state of the KRAS G12D mutant protein. Developed by Revolution Medicines, it functions as a "tri-complex" inhibitor that binds to the active form of the mutant KRAS protein to block downstream oncogenic signaling pathways. In preclinical models of pancreatic ductal adenocarcinoma (PDAC), RMC-9945 has demonstrated significant anti-tumor activity, both as a monotherapy and in combination with other agents. Notably, it has been shown to stimulate an interferon response and enhance the presentation of tumor antigens through increased MHC-I and MHC-II expression, which sensitizes tumors to T-cell-mediated immunotherapies and mRNA vaccines. It is frequently utilized in research as a tool compound representative of the clinical-stage investigational agent zoldonrasib.
