rmhTNF-NC is a recombinant mutant human tumor necrosis factor alpha (TNF-α) biologic drug developed as an immunotherapeutic agent. It is a structurally modified form of TNF-α with reduced toxicity (approximately five times less toxic and ten times more potent than wild-type TNF-α). The drug exerts antitumor effects through multiple mechanisms, including induction of apoptosis in tumor cells via mitochondrial and death receptor pathways, activation of inflammatory factors leading to vascular endothelial cell damage and thrombosis within tumors (causing ischemic necrosis), inhibition of neovascularization, modulation of immune function by enhancing T cell activity and cytokine production (such as interferon-γ and interleukin-2), promotion of neutrophil release from bone marrow, enhancement of phagocytic function, and stimulation of myeloperoxidase secretion. It is primarily used in combination with chemotherapy for refractory advanced non-small cell lung cancer (NSCLC) and non-Hodgkin’s lymphoma. Additionally, it is widely used locally to treat malignant pleural or peritoneal effusions associated with advanced cancers[1][3][4].
