Overview

RMP1-14 is a **rat IgG2a monoclonal antibody** that specifically targets the mouse **programmed death-1 (PD-1/CD279)** cell surface receptor, a member of the CD28 family encoded by the Pdcd1 gene[5][9]. This antibody binds to the extracellular domain of PD-1, blocking its interaction with the ligands PD-L1 and PD-L2, thus inhibiting the negative immune signal that normally leads to reduced T-cell activation and peripheral tolerance[1][3][9]. RMP1-14 is a standard reagent for *in vivo* murine experiments involving PD-1 pathway blockade and immune checkpoint research, widely used in preclinical models to assess immunotherapeutic strategies such as cancer immunotherapy or studies of autoimmune disease[1][2][6]. Its mechanism closely mirrors that of clinical anti-PD-1 therapies but is specifically designed for use in mouse and rat research settings[5][3]. Lower binding affinity compared to some alternatives like 1A12 is noted, requiring higher doses for effective PD-1 blockade in murine models[3].
