Overview

RMX2001 is an orally bioavailable, lipid-linked prodrug of the acyclic nucleoside phosphonate **9-(2-phosphonomethoxyethyl)guanine (PMEG)**. Developed by **Normax (Shanghai) Pharmaceutical Technology**, the compound is specifically designed for the treatment of **tuberculosis**, including multi-drug resistant (MDR-TB) and extensively drug-resistant (XDR-TB) strains. It utilizes hexadecyloxypropyl (HDP) lipid-conjugate technology to enhance oral bioavailability and facilitate the delivery of the active phosphonate into infected macrophages. Once intracellular, RMX2001 is metabolized into PMEG-diphosphate, which selectively inhibits mycobacterial **DNA-directed DNA polymerase** (specifically the DnaE1 subunit), leading to DNA chain termination and bacterial cell death. The drug has demonstrated potent in vitro and in vivo activity against *Mycobacterium tuberculosis* and various nontuberculous mycobacteria (NTM), representing a novel class of anti-tubercular agents with a mechanism distinct from current standard-of-care treatments.
