RN3161 is an investigational, GalNAc-conjugated small interfering RNA (siRNA) therapeutic developed by Rona Therapeutics to treat adults with overweight and obesity by targeting the liver-expressed gene INHBE, which encodes Activin E, a regulator of fat metabolism.[1][2][8][10][14] Using Rona’s GAIA siRNA and GalNAc hepatocyte-targeted delivery platform, RN3161 induces potent and durable INHBE mRNA silencing in hepatocytes, with preclinical monkey data showing over 90% knockdown after a low single dose and gene silencing lasting more than six months, enabling projected dosing every 6–12 months for long-term weight management.[1][2][6][8][10][14] By reducing Activin E signaling, RN3161 is intended to improve fat distribution, reduce adiposity while preserving lean mass, and enhance metabolic parameters such as lipids and glycemic risk markers, positioning it as a next-generation obesity therapy that may complement or follow GLP-1–based treatments.[1][2][8][10][14]
