Overview

RN6G is a humanized monoclonal antibody developed by Rinat Neuroscience Corporation (a research unit of Pfizer) for the treatment of geographic atrophy (GA) secondary to dry age-related macular degeneration (AMD)[1][2][5]. It binds with high affinity to the C-terminus of amyloid-beta (Aβ) peptides Aβ1-40 and Aβ1-42, which are implicated in the pathogenesis of both AMD and Alzheimer’s disease[1][4][5]. By binding these peptides, RN6G aims to reduce toxic amyloid deposits in retinal drusen and prevent their accumulation in the retina[3][7]. Unlike other anti-Aβ antibodies that target different regions or only one peptide form, RN6G targets both major forms and uses an IgG2Δa isotype designed to minimize immune effector functions for improved safety[1]. Despite promising preclinical results showing reduced amyloid plaque deposits in animal models, clinical trials were terminated due to lack of efficacy[3].
